An Ammonia-Induced Calcium Phosphate Nanostructure: A Potential Assay for Studying Osteoporosis and Bone Metastasis

چکیده

Osteoclastic resorption of bones plays a central role in both osteoporosis and bone metastasis. A reliable vitro assay that simulates osteoclastic vivo would significantly speed up the process developing effective therapeutic solutions for those diseases. Here, we reported development novel robust nanostructured calcium phosphate coating with unique functions on track-etched porous membrane by using an ammonia-induced mineralization (AiM) technique. The uniformly covers one side PET membrane, enabling testing resorption. pores allow mineral pins to grow inside, which, hand, enhances integration substrate and, other provides diffusion channels delivering drugs from lower chamber double-chamber cell culture system. applications processed were first demonstrated as drug screening device alendronate, widely used osteoporosis. It was confirmed delivery alendronate decreased number monocyte-differentiated osteoclasts To demonstrate application studying metastasis, delivered PC3 prostate cancer-conditioned medium differentiation monocytes into enhanced. This thus new platform activities assessing efficacy vitro.

Phosphate and carbonate salts of calcium support robust bone building in osteoporosis.

BACKGROUND Calcium is an essential cotherapy in osteoporosis treatment. The relative effectiveness of various calcium salts for this purpose is uncertain. Many older women with osteoporosis have phosphorus intakes of <70% of the Recommended Dietary Allowance. OBJECTIVE Our objective was to test the hypothesis that calcium phosphate would better support anabolic bone building than would calciu...

CALCIUM PHOSPHATE CEMENT: STUDY OF (BETA-TRICALCIUM PHOSPHATE, DICALCIUM PHOSPHATE AND CALCIUM CARBONATE SYSTEM

Calcium phosphate cements (CPCs), using B-tricalcium phosphate (ß-TCP, Ca3 (P04)2), dicalcium phosphate (DCP, CaHP04), calcium carbonate (Ca CO3), and hydroxylapatite (HAp, Ca10(P04)6(OH)2) as powder cement and disodium hydrogen phosphate (Na2HP04) solution as liquid component were prepared. After mixing the powder and liquid constituents, injectable and self-setting calcium phosphate cements (...

Calcium carbonate-calcium phosphate mixed cement compositions for bone reconstruction.

The feasibility of making calcium carbonate-calcium phosphate (CaCO(3)-CaP) mixed cements, comprising at least 40% (w/w) CaCO(3) in the dry powder ingredients, has been demonstrated. Several original cement compositions were obtained by mixing metastable crystalline CaCO(3) phases with metastable amorphous or crystalline CaP powders in aqueous medium. The cements set within at most 1 h at 37 de...

The incorporation of platelet-rich plasma into calcium phosphate cement enhances bone regeneration in osteoporosis.

BACKGROUND Polymethyl methacrylate (PMMA) bone cement is widely used for osteoplasty. However, previous studies have demonstrated the adverse effects of PMMA due to its excessive stiffness and heat production. Recently, calcium phosphate cement (CPC) that overcomes those negative effects has been successfully applied in osteoplasty. The potential problem of CPC is markedly less initial stiffnes...

New automated multiplex assay for bone turnover markers in osteoporosis.

BACKGROUND Serum C-terminal cross-linked telopeptide of type I collagen (CTX-I), N-terminal propeptide of type I collagen (PINP), and osteocalcin (OC) are among the most sensitive bone turnover markers for evaluating osteoporosis. Each marker is currently measured individually by manual or automated immunoassays that are time consuming and require substantial sample volume. We evaluated the per...
